Development of molecular carrier systems for selective thermal ablation and immunotherapy of metastases (SelekThermo)

The research project funded by the Society for Chemical Engineering and Biotechnology e.V. (DECHEMA) in collaboration with the Institute of Textile Machinery and High Performance Textile Technology (ITM) of the Technical University of Dresden, investigates novel nanocarriers produced by electrospinning. Such nanocarriers selectively target metastases and especially brain metastasis of renal cell and bladder carcinoma.

This is accomplished by arming the nanocarriers with single-chain antibodies or peptide ligands that exhibit specificity against prostate stem cell antigen (PSCA) or prostate-specific membrane antigen (PSMA) found in renal cell and bladder carcinoma tumors. For sustained anti-tumor effects, the nanocarrier system delivers Toll-like receptor agonists to induce a type I interferon immune response. To enhance an inflammatory cells death and improved release of tumor-antigens the nanocarriers contain superparamagnetic Fe3O4 crystals that are utilized for thermal ablation.

Thus, the objective of the project is to develop and validate highly selective and safe nanocarrier system for combined immunotherapy and thermal ablation of metastases and brain metastases.
