Superparamagnetic nanoparticles for the diagnosis and thermal ablation of prostate stem cell antigen (PSCA)-positive tumors and brain metastases of small cell renal cell carcinoma

The project, which is performed in close collaboration with the Urological Research Laboratory of the University Hospital Carl Gustav Carus (Head Prof. Dr. Füssel) deals with an experimental thermoablation approach to selectively treat tumors and brain metastases of renal cell carcinoma (RCC). For this, supraparamagnetic iron oxide nanoparticles (SPION) are utilized. Current therapeutic concepts are limited by the development of tumor resistance mechanisms, whereas the use of SPIONs offers a promising treatment option. Such particles can be stimulated to produce heat by applying an external alternating magnetic field. Temperatures above 42°C induce apoptosis or coagulation necrosis in the tumor tissue. The untreated, healthy tissue is not damaged in this process, since the influence of the used field strengths is negligible. The objective of this project is the development and functional testing of a targeted delivery of SPIONs to tumor cells using, single chain antibodies (scFv). For this a recently established humanized anti-PSCA-scFv which specifically binds to PSCA-positive tumors of RCC will be employed. Assembly of scFv and SPIONs will be performed using a convenient streptavidin-biotin system. The selective uptake and experimental thermoablation will be investigated using PSCA-positive tumor cells as well as primary cells from RCC brain metastases.
